The canonical role for RNA is an intermediate between genes and proteins, but we are increasingly finding that RNA can itself regulate whether and how much genes are transcribed. After foraying into the world of RNA sequencing, I realized there was a wealth of information contained in sequencing data with very little mechanistic understanding about what up or downregulation of a particular RNA transcript does. This sent me diving into the fascinating rabbit hole that is RNA biology.<br><br>Through this experience, I hope to learn how to operate independently as a scientist. I see doing research as an opportunity to affect positive change in the world. With that opportunity comes the imperative to pursue the truth to the best of our abilities. It turns out that pursuing the truth to the best of my abilities is challenging in innumerous ways, and a PhD is preparing me for those challenges. I think doing a PhD has also allowed me to develop several ‘soft skills’ important to doing research, including critical thinking and dealing with uncertainty and failure. Beyond soft skills, I’ve gained so much knowledge and technical expertise in the field of biochemistry that I never would have otherwise. Taking core courses, taking the written and oral exams, and writing and defending proposals have all forced me to confront gaps in my knowledge and grow as a scientist.<br>&nbsp;</p><h5>WHERE I'M HEADED POST DOCTORAL STUDIES</h5><p>My undergraduate degree and research was in the field of neuroscience, and I’m still very much interested in neuroscience questions. I think I would like to apply the techniques and principles I’m learning to questions related to human neurological health and disease. With an organ as complex and important as the brain, regulation of the biochemical activities within brain cell types is likely to be specialized. Indeed, many non-coding mutations (and other likely RNA-driven phenotypes) cause an array of neurodevelopmental symptoms. Having gained an understanding of biochemical techniques and the challenges of implementing them for neuroscience questions, I hope to be able to design biochemical experiments that reveal truths pertinent to neurobiology.<br>&nbsp;</p><h5>BIOCHEMISTRY INSPIRATION</h5><p>It feels like a salient time for the marriage of basic biology and chemistry with advancements in human health. I’m inspired by the possibilities presented by gene editing technologies which can eventually be utilized for gene therapies. I’m inspired by the development of super-resolution imaging techniques that allow us to see single molecules in live cells and single atoms in biomolecules. I’m inspired by huge advancements in in silico techniques (including alpha-fold, and I’m hopeful that a similar algorithm will one day be able to predict RNA structure). More than anything, though, I’m inspired by biochemists—their expertise, creativity, and desire to improve our society.<br>&nbsp;</p><h5>WHY I SELECTED CU BOULDER</h5><p>When deciding which graduate programs to apply to, I tried to wholistically consider my experience in that environment both inside and outside of the lab. I’m very grounded by being surrounded by nature, so I knew CU ֲý had to be in the running as one of the most beautiful locales in the country. When I interviewed, I gleaned a culture of collegiality in the department that is pervasive from faculty to graduate students. The other graduate students seemed like a group I would fit with. I love that they take the science seriously but also pursue extracurricular hobbies and care about broader social issues. Scientifically, I wanted to choose a graduate program that would challenge me and push me to grow. I was aware of the Biochemistry Department’s reputation for highly rigorous and quantitative science. On top of that, the department is world renowned for basic RNA biology, an area I was interested to delve into.<br>&nbsp;</p><h5>EXTRA-CURRICULAR ACTIVITIES</h5><p>One important part of my graduate school journey has been serving on the board of Science Community Outreach Program and Education (SCOPE), an outreach organization that brings experiments and volunteer scientists to local middle schools. Being given the opportunity to do research as a young person was one of the most formative experiences of my life, and SCOPE’s mission is to give that experience to as many kids as possible by introducing them to hands-on science. I have served as SCOPE’s Volunteer Coordinator, Vice President, and Fundraising Coordinator in addition to serving as a volunteer and lesson lead in classrooms. This experience has taught me so much about leadership and education and solidified my desire to pay forward the excellent mentorship I have received throughout my career. I became interested in this topic because there were many unanswered questions about mitochondrial DNA organization, despite this process being extremely important for overall cell health. I was especially interested in this system because many unknowns could be answered using simple biochemical methods. I showed that TFAM, the protein responsible for packaging mitochondrial DNA, forms homogenous higher order complexes on DNA that exhibit microscale conformational dynamics. These dynamics may be a method of regulating access to the mitochondrial genome which is essential for cellular energy production.</p><p><br>I also worked on two other projects investigating histone-mediated genome organization in bacteria and giant viruses. In this work, I characterized three different mechanisms of DNA packaging, highlighting the diverse strategies adopted by these small but versatile proteins. Together, my work in graduate school characterized the fundamental process of genome organization across the domains of life, contributing to the quest to understand the origins of the eukaryotic nucleus. I hope that I leave this experience as a highly skilling, rigorous scientist that is an expert in my field.</p><h4><br>WHERE I’M HEADED POST DOCTORAL STUDIES</h4><p><br>I anticipate changes to my research focus as I move forward. I would like to learn cryogenic electron tomography to expand my skillset as a structural biologist and to study cellular processes in their native state. I’m excited to explore new organisms and mechanisms that I’m yet to come across; there are so many interesting questions out there.</p><h4><br>WHY I SELECTED CU BOULDER</h4><p><br>I chose the CU ֲý Department of Biochemistry for graduate school because of its collaborative culture, the well-equipped facilities available to students and the many highly active research labs. Even though I sat toward the back and was told I talked too much by the teachers in those classes, I found myself drawn to the nature of systematic problem solving and testing inherent in the field. So, knowing that I wanted to pursue something in the area of biochemistry or molecular biology and being lucky enough to be awarded the Boettcher scholarship, I chose to come to CU for my degree and to hopefully gain experience that would clarify how I wanted to use science in my career.</p><h5><br>BIOCHEMISTRY AS INSPIRATION</h5><p><span>I didn't have a set plan when starting at CU for exactly what I wanted to do. Knowing I liked science and was okay in the classes meant that I thought about a PhD as an option. I also grew up in a family where there were some physicians so that was also in--&gt; on my mind as a possibility. So, in addition to time in the lab, I was able to get some experiences in healthcare volunteer work and shadowing, which I enjoyed but were not overwhelmingly convincing to me for a career as a doctor. What I, for better or worse, found most compelling to choosing a direction during college was the experience of my sister getting sick and my family and personal feelings of helplessness both from a medical and systems perspective. So there was a lot of meaning for me created around the idea of being able to alleviate these feelings of powerlessness. And how could the more direct practice of scientific and biochemical research fit into that? I think what took me a long time (perhaps too long) to fully appreciate was how the scientific discovery and story I worked on in the lab more broadly could be applied to treatment of diseases like cancer or more rare orphan conditions that didn't have treatment options currently available. So ultimately I became most interested in how fundamental discoveries could be translated to the clinical space.</span><br>&nbsp;</p><h5>CONNECTIONS MAKE A DIFFERENCE: GAINING REAL-WORLD EXPERIENCE</h5><p><span>A family friend had happened to become an acquaintance with </span><a href="/lab/cech/" rel="nofollow"><span>Dr. Thomas R. Cech</span></a><span> while he was at HHMI and suggested I look into working in his lab as a form of practical experience when starting my studies. So as a freshman I started working in the lab, dutifully making plates, cell culture media, and buffers, and timidly using the autoclave. Over time I started working with one of the postdoctoral fellows, learning how to perform site directed mutagenesis of plasmids, transforming bacteria, and generating sizeable amounts of mutated enzyme for a variety of assays. A screening process now referred to by Dr. Cech as "obsolete" and "we would never do this now." But the exercise of getting to take exceptional chemistry and biochemistry courses from faculty, paired with the real-world experience of generating hypotheses and designing and running experiments to test relevant and important scientific questions, allowed me to gain a broad appreciation of the field. With more time I was actually able to meaningfully contribute effort and usable data to his project, understanding how multiple years of work and many experimental iterations came together to ultimately form a full scientific story. This is the story that was ultimately published in Nature and served as the basis for my honors thesis, my first real exercise in scientific writing. Others credited from CU ֲý include: Nandakumar J (this was the amazing postdoc I worked with, now on faculty at Michigan), Weidenfeld I, Zaug AJ, Leinwand LA, Cech TR.</span></p><p class="text-align-center"><a class="ucb-link-button ucb-link-button-gold ucb-link-button-default ucb-link-button-regular" href="https://pubmed.ncbi.nlm.nih.gov/23103865/" rel="nofollow"><span class="ucb-link-button-contents">First Published Paper - Nature 2012 Dec 13</span></a><br>&nbsp;</p><h5>COMING FULL CIRCLE FOLLOWING CU UNDERGRADUATE PROGRAM: ASSISTANT PROFESSOR OF CARDIOLOGY - CU ANSCHUTZ</h5><p>So, after a lot more school and a lot more training at different places across the country (<strong>Vanderbilt, Dana Farber, Stanford</strong>) with that ongoing--&gt; an ongoing interest in scientific and medical research, I'm now back home in Colorado with an actual job as an Assistant Professor of Cardiology at the medical center. I have developed an expertise in an area of medicine called cardio-oncology where I see patients who have experienced cardiotoxic side effects from their cancer therapy, have cardiovascular disease that needs management during their cancer therapy, or cancer survivors looking for guidance on cardiovascular health. This helps to inform my research program that works to understand how and why cancer patients have worse cardiovascular disease even after cure, and ways we can use new and existing therapies to change this. This has allowed me to keep working in basic and translational science, pilot and Phase 1 clinical trial work, and preclinical and clinical biomedical industry consulting work. Though I draw on a lot of different training experiences I've had in my current career, there are&nbsp;fundamentals I learned while at CU that I use every day whether that is troubleshooting a basic assay in the lab with my technician, thinking critically about optimal experimental design, assessing important literature coming out in the field, or thinking about how to best train and prepare future physicians and scientists.<br>&nbsp;</p><h5>ON BEING SUCCESSFUL IN BIOCHEMISTRY</h5><blockquote><p>Consistently showing up and doing your best is a large portion of the battle. Working in the Cech lab was one of the most formative experiences for me while in college, and it was later revealed to me that half of why I was able to spend a full 4 years there was the fact that I kept showing up and reliably working hard. The other half was that Dr. Cech had forgotten to ask me to stop coming, his words.<br><br>Stay flexible, open minded, and reflective, even if you think you have it all planned out. You have likely had and will continue to have experiences that change your perspectives or motivations.</p><p>Working in science and research is a team sport; use your time at CU to help think about what excites you about that team sport and what skills you want to bring to the table going forward. I chose to pursue my undergraduate studies at the ֲý because of my love for the state of Colorado and the strength of its close-knit yet accomplished Department of Biochemistry. As an R1 research institution, CU ֲý offered me the opportunity to engage in meaningful research early in my academic career, which was also a key component in my decision.<br><br>I began my research career in 2022 in the laboratory of Dr. Jeffrey Cameron, where I studied the life-sustaining characteristics of cyanobacteria using inducible CRISPRi methodologies. Following Dr. Cameron’s passing in 2024, I continued my research training in the laboratory of Dr. Karolin Luger, where I investigated interactions between the transcriptional cofactor TRIM28 and chromatin. My research interests include chromatin biochemistry, structural biology, drug development, and cancer research.</p><h5><br>BIOCHEMISTRY AS INSPIRATION</h5><p>I am fundamentally inspired by the interconnectedness of molecular systems and biochemical pathways. I find it intriguing to problem solve how these two concepts are related. A unique quality to me is my interest to view science through an artistic creative lens, specifically, my notes are filled with hand-drawn molecular diagrams that help me connect artistic principles with biochemical concepts. This interrelationship between art and science for me has defined my individual approach to biochemical research.<br><br>My journey as a scientist has been shaped by Dr. Cameron, whose mentorship and passion for research continues to influence my academic path and personal interests. I have also received UROP funding for my research for the past three years.</p><h5><br>CHARTING MY OWN PATH: CU BIOCHEMISTRY + GRADUATE STUDIES</h5><p>I hope my college experience in biochemistry will give me a deeper understanding of the physical mechanisms underlying the natural world, allowing me to explore possible therapeutics to treat disease. I am interested in bringing an appreciation for the artistic and creative aspects of biochemistry into my graduate studies.<br><br>As I enter graduate school, I look forward to exploring diverse areas within structural biology. I developed my interest in cancer research, cell signaling and systems biology before coming to grad school. My time working&nbsp;in industry research (AbbVie - formerly Stemcentrx) really solidified my love of science, helped&nbsp;focus my research interests, and better prepared me for grad school.<br>&nbsp;</p><p>Being selected in 2021-2023 (renewed in 2025) to participate as a trainee on the NIH sponsored T32&nbsp;Graduate Training Program in Signaling and Cellular Regulation has been a great experience. I&nbsp;really appreciated the cross-disciplinary approach and feel it fosters an interdepartmental sense&nbsp;of community, building connections between the Biochemistry and the Molecular, Cellular &amp;&nbsp;Developmental Biology departments at CU ֲý.</p><h5>WHERE I’M HEADED NOW</h5><p>I hope my grad school training will broaden my scientific understanding to better develop a&nbsp;scientific question and think critically. I love the problem-solving aspect of research, being able&nbsp;to troubleshoot and design my own experiments. I’ve gained more computational skills during my&nbsp;graduate training that I hope to continue using and expand on in my future career.</p><p>After graduating, I hope to continue studying cancer research and help develop therapeutics in an&nbsp;industry setting. After getting my undergraduate degree, I&nbsp;lived in South Korea for a year teaching English. During this time, I experienced a lot of personal&nbsp;growth but ultimately realized I missed science. Afterwards, I worked in several different biotech&nbsp;companies in the SF Bay Area. While I gained a lot of important skills, I realized I wanted to better contribute and guide our research goals. Outside of working on a double major (Biochemistry with the recent addition of Chemistry), Asher is Vice President of the <em>Science Community Outreach Program and Education (S.C.O.P.E.</em>) program, which aims&nbsp;to increase science identity and access for middle school students. He is also Vice President of the <em>CU Trampoline Club... n</em>ow you understand why we know he has "boundless energy"!</p><p><strong>CU BOULDER = THE PLACE FOR ME</strong><br>I chose to do biochemistry at CU ֲý for several reasons. One is the abundance of world class research all set against a backdrop of beautiful mountains. Another is the culture of supporting undergraduate research endeavors and the accessibility of laboratory research. Also, as someone who wants to pursue a career in biotech, having a designated biotechnology building with 3 floors of cutting-edge research labs was a big plus.&nbsp;</p><p>I just declared a major in chemistry in addition to biochemistry! I recently realized that I can’t feel as confident in my knowledge if I don’t have a deeper understanding of the latter half of biochemistry. Also, I found that I really enjoy organic chemistry and after my experience researching metalloenzymes at MIT this summer I have become fascinated with bio inorganic chemistry. I’m taking this next semester as a sort of trial run to see how much I enjoy pure chemistry and the corresponding bench work.</p><p><strong>UNDERGRADUATE RESEARCH OPPORTUNITY: INVALUABLE KNOWLEDGE&nbsp;</strong><br>I hope a biochemistry degree gives me the toolset and confidence to propose solutions to complex biochemical problems and invent new assays and methods for discovery. I think what this program has already allowed me to do in two years is incredible and makes me excited for the latter half of my college experience.</p><p>I believe living matter is the most fascinating phenomenon in the universe. It’s endlessly complex and adaptive: it can facilitate novel chemical reactions and survive across an enormous range of environments. Biochemistry is life at the smallest possible scale and is therefore the root cause of all diseases and afflictions. The sheer complexity of life at its most basic components means there’s so much space to invent and discover. Part of why I want to fight disease so much is because I know how it feels to wake up one day and have your entire world be turned upside down by a biochemical phenomenon completely outside of your control.</p><p>&nbsp;</p><p>&nbsp;</p><p><strong>CHARTING MY OWN PATH: CU BIOCHEMISTRY SUPPORT</strong><br>At the end of the spring semester, I reached out to Dr. Catherine Drennan at MIT with my CV and a cover letter to apply to work in her lab that summer. Coincidentally, HHMI reinstated its <em>Summer Undergraduate Research Program (HHMI SURP) </em>at the same time, so she nominated me for it.&nbsp;</p><p>This couldn’t have happened without my current PI at CU ֲý, Dr. Karolin Luger, vouching for me.</p><p>I’m from Boston so I biked to MIT every day from home which was really nice. I really appreciated how interdisciplinary the lab was, I met grad students and post docs with backgrounds in organic, inorganic, and biochemistry - all fields I’m interested in exploring! I enjoyed my independence this summer; I got to design and conduct my own experiments as well as invent my own methods. My big takeaways from this experience are: science should always be done creatively, and a positive work environment and culture is the most important criterion for choosing a lab.</p><p><strong>MY GRATITUDE AND FORWARD-THINKING</strong></p><blockquote><p><em>“I am really honored to have been awarded the Bob and Dickie Lacher scholarship in AY2024-2025 and again this academic year. And... he did all of this while earning top honors - both in academic achievements and recognition from faculty and peers alike. Let's see what his path looked like!</p><h4><strong>CU BOULDER - A PLACE THAT FEELS LIKE HOME</strong></h4><p>I chose to attend CU ֲý because I grew up in Colorado, graduating from Gilpin County Undivided High School, and loved the atmosphere, the beautiful campus, and the opportunity to be able to explore options outside of my major. CU offers programs in pretty much anything you could ever want to do, and I actually declared a second major in Classical Studies and a minor in Hebrew/Israel Studies since beginning my studies here. There aren’t a lot of schools that can offer that and I am super happy to have been able to study a broad range of subjects that I was interested in. I didn’t know much about biochemistry or the department here at CU, I just liked biology and chemistry in high school. However, after taking BCHM classes, meeting professors, and conducting research here, I absolutely love the program. I think that the faculty are very knowledgeable, accomplished, and friendly, and the facilities for the BCHM department are excellent.</p><h4><br><strong>UNDERGRADUATE RESEARCH OPPORTUNITY = INVALUABLE KNOWLEDGE</strong></h4><p>I think that the biggest source of inspiration for me in Biochemistry is the amount that we don’t know. I am absolutely fascinated by the mechanisms, structures, and pathways that we already know, and learning those is super fun for me. However, in class or in the research lab, I am constantly reminded of all of the things we don’t know. I also think that research has been a huge component of my time here at CU. I started in an organic chemistry laboratory in the Spring 2023 semester and worked there through the summer, transitioning to my current biochemistry laboratory in Fall 2023. I am super lucky and thankful to have been working on research in the Luger Lab since then, and I am eternally grateful to the Luger Lab and my graduate student mentor for the opportunity and guidance in developing valuable lab skills and gaining experience in the field of biochemical research. Currently, I am hoping to take a gap year and work at a biotech or related company to learn more about non-academic research. I am undecided on whether or not I will be attending graduate school, but I believe that my college experience in Biochemistry will allow me to contribute to the scientific community and give me the tools I need to pursue whatever opportunities, careers, or further interests I may have.</p><p>&nbsp;</p><h4><strong>In Academics</strong></h4><p>In Damian's pursuit of a BA in Biochemistry, he was named this year's American Institute of Chemists Student Award (as voted by faculty). Starting in Spring 2023 and continuing through this spring (2025) he was on the Arts and Sciences Dean's List. The strength of academic programs offered here plus the beauty of ֲý were top of Mila's list - plus - the tennis team wanted her. She has continued to be a stand-out star - both on the court and in the academic research lab.</p><h4><strong>Heidelberg, Germany to CU ֲý: A Fruitful Choice</strong></h4><p>Born and raised in Germany before moving to ֲý for college, Mila's journey as an<br>international student-athlete at CU has been incredibly rewarding. She selected the Department of Biochemistry at CU ֲý for its strong faculty - giving her the opportunity to learn from amazing professors who are not only leaders in the Biochemistry field but also care for their students’ success. She attributes their passion for teaching and mentorship to creating a great learning environment where she personally felt encouraged to grow her scientific understanding.</p><p>Their support has played a crucial role in shaping her academic journey and sparking her curiosity for biochemistry. In addition, the department offers great undergraduate research opportunities where she feels very fortunate to join Dr. Roy Parker’s lab. Individual success and development are strongly supported in the department of Biochemistry, which she says has helped her become a better scientist.</p><h4><strong>Undergraduate Research Opportunity: Invaluable Mentorship</strong></h4><p>Mila knew, going into her senior year of college, that she wanted to gain more research experience and sought out an opportunity to join a lab for a summer internship. In Dr. Parker’s lab her research focus is looking at a protein called G3BP1, which is an essential regulator of cellular RNA metabolism. The main known functions of G3BP1 are regulating RNA stability and translation in the cytoplasm. Upon stress, G3BP1 also forms cytoplasmic stress granules, a type of membranelles assembly that forms through the biophysical process of condensation. Her main goal is to understand the mechanism of G3BP1 nuclear- cytoplasmic localization in the cell.</p><p>She spends 40 hours per week working in the lab during the summer and while it is a lot of hard work spent in the lab it is also the most fun experience at the same time. One of the biggest reasons she spends so much time in the Parker lab is the incredible mentorship she is given. The guidance and support from experienced researchers created an environment where she felt encouraged to ask questions, think critically, and push herself to improve. Learning something new each day, whether it was troubleshooting an experiment, analyzing data, or perfecting a new technique, made the experience truly enriching. This experience made it clear that she wanted to pursue a career in research.</p><div class="row ucb-column-container"><div class="col ucb-column"><h4><strong>Blending Success in an Individual Sport into Athletic and Scientific Teamwork</strong></h4><p>Mila was motivated to find a strong team environment where she has the opportunity to learn from the people around her. Engaging with such a diverse and driven group has taught her the value of collaboration, support and teamwork. Coming from an individual sport, her focus was always on personal success and by transitioning to college athletics, where teamwork is essential, she has found an incredibly valuable experience. She realizes that this lesson translates seamlessly into the research setting where, while each person works on their own project, the true strength of a lab comes from an environment built on teamwork, respect, and mutual support. As a Chicana, I recognize the barriers imposed upon disadvantaged groups and know that I would not be where I am today without the aid of programs that increase support for diverse students in science. I felt a responsibility to give back to the same community that helped me to succeed as a young researcher. So, I have been involved in various outreach and mentoring programs such as the Society for the Advancement of Chicano and Native Americans in STEM (SACNAS), Peer-2-Peer mentoring, Summer Multicultural Access to Research Training Program (SMART), Longmont High School SMART Program, STEM Routes, and the Uplift Program. I also co-founded and was the former chair of the Biochemistry Department’s DEI committee. Outreach work is what can inspire the next generation of scientists and it is critical that we create an inclusive environment for all identities to be able to thrive.&nbsp;</p><p>&nbsp;</p><h3>My Research: Biochemistry for Creating New Tools for Human Health</h3><p>My research focuses on elucidating the role of biomolecular condensates in transcription regulation. Biomolecular condensates are membraneless liquid droplets that possess a high concentration of local proteins and/or nucleic acids. These condensates offer spatiotemporal control of protein function and could be utilized by the cell to regulate processes such as transcription. Specifically, most of my efforts have focused on developing a fluorescent assay to simultaneously visualize in real-time condensate formation and human RNA polymerase II transcription using a defined reconstituted in vitro system. I knew this project was a great fit for my thesis research because it combined researching the regulation of a fundamental process with the development of a new method or tool. Additionally, the implication of biomolecular condensates in coordinating transcription is a relatively new idea and subsequently, there are a lot of available questions that need to be addressed so we can better understand the underlying mechanism. Finally, I also find studying human transcription intriguing. It a complex orchestration of a multitude of proteins and nucleic acids where the precise timing is critical for proper gene expression.</p><p>&nbsp;</p><h3>My Life's Journey</h3><p>Biochemistry truly fascinates me, every organism surrounding us survives because of all these thousands of intricate biochemical interactions. I am inspired by the idea that I could be someone who contributes to understanding how these interactions regulate processes that give rise to life. I chose CU ֲý because it felt like an environment where I could succeed. Graduate students here appeared to be genuinely happy. They were working hard in lab and simultaneously enjoying their weekends hiking or spending time with friends. These students were publishing in top journals and still finding time to enjoy life. That dynamic is exactly what I wanted in a program. Additionally, the research in the biochemistry department and the CU community is astounding and the faculty are easy to talk to despite their impressive backgrounds.
